Creatine kinase has four kinds of isoenzyme: muscle (MM), brain (BB), hybrid (MB) and mitochondrial (MiMi) and mainly exist in muscle cells, brain cells, myocardial cells, and the mitochondria of myocardial and skeletal muscle cells, respectively. Creatine kinase isoenzyme plays a significant role in a variety of diseases. The creatine kinase level increase rapidly in the serum when experiencing muscle atrophy, myocardial infarction and other pathologies. Thus, it is now believed that the determination of creatine kinase activity is more reliable than the electrocardiogram in the diagnosis of myocardial infarction. The creatine kinase isoenzyme (CK-MB) has the highest specificity among the other creatine kinase. Creatine kinase has attracted extensive attention and in-depth research because of its important physiological function and clinical value.||Intended Use:|The kit is applicable to colorimetric determination for ultraviolet spectrophotometer or automatic biochemical analyzer.||Range: |0-200U/L, r2≥0.995||Test Principle:|Creatine kinase (CK) has three kinds of dimers composed of subunits of M and B. CK1 (CK), CK2 (MB) and CK3 (MM) are mainly found in the cytoplasm. There were also a small amount of Mitochondrial isozymes (CK-MiMi) in the serum. In the myocardium, CK activity is high (only lower than the activity in skeletal muscle), and CK-MB accounts for 13% to 22% (<1% in the skeletal muscle). Therefore, CK- MB is a specific indicator of myocardial injury. Immunosuppression method, which is adopted in this kit, inhibits M subunit with anti-M serum to determine the activity of the remaining B subunit. The activity of CK-MB can be calculated by doubling the activity of the remaining B subunit.||Accuracy: |Inaccuracy≤15%||Precision: |Intra-Assay CV: <6%|Inter-Assay CV: <15%||Kit Components (see Protocol for final kit quantities):|Reagent 1,|Imidazole buffer (pH6.6), 100×10-3mol/L|Glucose, 20×10-3mol/L|Magnesium acetate, 10×10-3mol/L|Ethylenediaminetetraacetic acid, 2×10-3mol/L|Hexokinase (HK), 3000U/L|Adenosine diphosphate (ADP), 2×10-3mol/L|Adenosine monophosphate, 5×10-3mol/L|Two adenosine phosphate, 10×10-6mol/L|N-acetylcysteine, 20×10-3mol/L|Oxidized coenzyme II (NADP+), 2×10-3mol/L|Anti-CK-M antibody, 2000U/L|Glucose-6-phosphate dehydrogenase (G-6PDH), 2500U/L||Reagent 2,|Creatine phosphate, 30×10-3mol/L||Storage and Stability:|Store powder at 4°C liquid at -20°C.
