ELISA Kit Principle | Aviva Systems Biology GABA ELISA Kit (OKEH02564) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-GABA antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated GABA and incubated. The GABA found in the sample or standards competes with the biotinylated GABA for limited binding sites on the immobilized anti-GABA antibody. Excess unbound biotinylated GABA and sample or standard GABA is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated GABA captured in the well and inversely proportional to the amount of GABA which was contained in the sample or standard. | ||||||||||||||||||||||

Additional Information | Function: g-Aminobutyric acid is the chief inhibitory neurotransmitter in the mammalian central nervous system. It plays a role in regulating neuronal excitability throughout the nervous system. In humans, GABA is also directly responsible for the regulation of muscle tone. Although chemically it is an amino acid, GABA is rarely referred to as such in the scientific or medical communities, because the term "amino acid," used without a qualifier, conventionally refers to the alpha amino acids, which GABA is not, nor is it ever incorporated into a protein. In spastic diplegia in humans, GABA absorption becomes impaired by nerves damaged from the condition's upper motor neuron lesion, which leads to hypertonia of the muscles signaled by those nerves that can no longer absorb GABA. | ||||||||||||||||||||||
